RICTOR, is a key component of the mTOR complex 2 (mTORC2) and is required for phosphorylation of Akt at serine 473.Rictor is overexpressed in several cancers leading to hyperactive mTORC2 and has been shown to play a causal role in glioma formation. The Rictor-FRAP complex plays a role in PKCα phosphorylation, directly phosphorylates Akt/PKB on Ser 473 in vitro and facilitates Thr 308 phosphorylation by PDK1. It also may function as a drug target in tumors that have lost expression of PTEN, a tumor suppressor that opposes activation of Akt/PKB.
