GLI1 (Glioma associated oncogene 1) is a member of the GLI C ₂ H ₂ - type zinc finger protein family. At rest, GLI1 is anchored in the cytoplasm by binding to the SUFU (Suppressor of Fused) protein. When the signal is activated, GLI1 undergoes conformational changes through its interaction with STK36 (Serine/Tetranine Kinase 36), thereby releasing SUFU inhibition and transferring to the nucleus. In addition, ULK3 (Unc-51 Like Autophagy Activating Kinase 3) mediated phosphorylation modification has been shown to promote nuclear localization of GLI1. As a transcription activator, GLI1 regulates the expression of downstream genes by recognizing and binding to the DNA consensus sequence 5 '- GACCCCA-3'. GLI1 can mediate SHH signaling and play a role in cell proliferation and differentiation through its role in SHH signaling. GLI1 can also promote cancer cell migration.
